2002, GastroenterologyCitation Excerpt :The corporal biopsy specimens were taken in a preset order: at the NIH, 4 specimens were taken from the greater curvature (3 at La Sapienza) and 4 from the lesser curvature (3 at La Sapienza).39,40 Biopsy specimens were fixed for 3–5 hours in Bouin's solution, rinsed with 70% ethyl alcohol and 30% water, and processed as described previously.39 Serial 5-μm-thick sections of gastric body mucosa perpendicular to the mucosal surface were stained with H&E for evaluation of the type of mucosa, with Giemsa stain for H. pylori evaluation, and with immunostaining for endocrine cells by using (1) monoclonal antibody against chromogranin A (clone LK2H10; Biogenex Laboratories, San Ramon, CA; working dilution 1:250) and (2) polyclonal rabbit antisera against the α-subunit of human chorionic gonadotropin (α-hCG; produced by Dr. H. Okumura, Teikoku Hormone Manufacturing Co., Kawasaki, Japan, donated by Dr. M. Fukayama, Department of Pathology, Tokyo Metropolitan Komagone Hospital, Tokyo, Japan; diluted 1:1600) as primary antibodies, with overnight incubation at 4°C.
